PHARMACOLOGICAL PROPERTIES

ATC vet classification code QJ01 – Antibacterial veterinary drugs for systemic use. QJ01FA10 – Azithromycin.

Azithromycin is a macrolide antibiotic belonging to the azalide group. The molecule is formed by introducing a nitrogen atom into the lactone ring of erythromycin A. Azithromycin's mechanism of action is the inhibition of bacterial protein synthesis by binding to the 50S ribosomal subunit and preventing peptide translocation.

Azithromycin exhibits a post-antibiotic effect. This effect is based on irreversible changes in the microorganism’s ribosomes, leading to the persistent blockade of protein translocation. As a result, the overall antibacterial effect of the drug is enhanced and prolonged, remaining effective for the time necessary for the synthesis of new functional proteins in the microbial cell.

Azithromycin has a broad antimicrobial spectrum against Gram-positive and Gram-negative bacteria (such as Actinobacillus lignieresi, Haemophilus spp., Moraxella spp., Bordetella spp., Campylobacter spp., Legionella pneumophila, Salmonella spp., Escherichia spp., Listeria spp., Staphylococcus aureus, Staphylococcus spp., Streptococcus spp., S. pneumoniae, S. pyogenes, Enterococcus faecalis, Erysipelothrix insidiosa), some anaerobic bacteria (Clostridium perfringens, Fusobacterium spp., Pasteurella (Mannheimia) haemolytica, P. multocida, Haemophilus parasuis), mycoplasms (Mycoplasma pneumonia), chlamydia (Chlamydia pneumonia), and spirochetes (Borrelia spp.).

When administered orally, azithromycin is well absorbed and rapidly distributed in the body tissues, reaching high concentrations that exceed plasma concentrations. The high antibacterial activity is achieved due to azithromycin’s ability to penetrate and accumulate intracellularly in leukocytes, which transport the antibiotic to the sites of inflammation, leading to a concentration of the drug being six times higher at the site of inflammation compared to intact tissues.

The high concentration remains for 5-7 days after the last dose of azithromycin.

More than 50% of the dose is excreted unchanged from the body, primarily through bile and partially through urine.

INDICATIONS FOR USE

Treatment of dogs and pigeons with respiratory diseases, gastrointestinal disorders, urinary system infections, mixed and secondary infections during viral diseases, caused by microorganisms sensitive to azithromycin.

DOSAGE AND ADMINISTRATION

Administer orally with drinking water or food in the following doses:

Pigeons – 1 g of the product per 3-6 L of drinking water, daily, for 3-5 days.

Dogs – 0.25-0.5 g of the product per 10 kg of body weight, daily, for 5-7 days.

CONTRAINDICATIONS

Hypersensitivity to the components of the product, or to other antibiotics of the macrolide/ketolide group.

Not for use in animals with severe liver or kidney dysfunction, pronounced bradycardia, arrhythmia, or severe heart failure.

Not for use in pregnant animals.

Not for use with heparin.

Not for use in productive animals!

CAUTIONS

Tetracyclines and chloramphenicol enhance the effect of azithromycin (synergism).

Lincosamides reduce the effect of azithromycin.

When used at recommended doses, side effects are generally not observed. In case of side effects (itching, nausea, vomiting, muscle tremor), stop using the product and provide desensitizing therapy.

The product should only be used when it is clear that the therapeutic benefit outweighs the potential negative consequences.

In case of overdose, animals may experience restlessness, disorientation, temporary hearing loss, and increased liver enzyme levels in the blood serum. In such cases, discontinuation of the product is recommended.
